I'm trying to find out which lentiviral shRNA system is best for me.

I have 1~3 genes to silence (alone or simultaneously). However, siRNA transfection by lipid-based reagent already affected my target genes. So I would like to use viral-based strategy, and I can play it with stable silencing.

Is there any lentiviral system that meets the following requirments?

1. Promoter should be inducible by BBB-pentrated drugs like DOX, for I am doing brain tumor research.

2. Infused with fluorescent protein like GFP or RFP, so I can do sorting rather than antibiotic selection, for the sake of cell viability.

3. Produce high titer virus so I do not have to work on the painful purification stuff.

4. The basic plasmid can express in a mammalian system, so I can do shRNA segments evaluation easily and quickly in 293 cells, rather than packing an intact lentivirus for selection.

Besides, where can I find a reliable software or algorithm to design my own shRNA, or an reliable shRNA database where the segments have been validated?
