I want to synthesize RACE cDNA from wheat RNA. I have experience in RACE and use SMART kit from Clontech and FirstChoice® RLM-RACE from life tech (2 years back) and found SMART much better. Can anyone suggest the most successful kit for RACE cDNA synthesis this time?
I've used Clontech's kit and the one from Life Technologies, and I personally found Life's much easier to work with (and with good results). Price? also expensive, but it is worth the price.
The basic concepts of SMART kit and RLM-RACE are slightly different. RLM-based RACEs involve in CIP and TAP treatments, which only select complete mRNA messages ('true' messages) by chasing 5'-cap. If you want to identify the exact base-pair position of transcription, such as identifying a transcription start site, using RLM-based RACEs is a 'must' for accurate predictions, but those kits are usually a bit more expensive. I have never used one, but people recommended for GeneRacer RACE kit from Invitrogen/Life technologies.
If you just want to clone a whole cDNA (and ignore clones that contain incomplete messages, i.e., degraded messages), SMARTer kit from Clontech will do the job nicely. SMARTer kit is better than SMART kit as Andrei mentioned.

I have experience of working with invitrogen RACE kits. For 5' RACE, clonetch kits yield better results.
Invitrogen's RLM Race kit has not given me good results with 5' or 3' race. I am lookign at ordering roche or clonetech's race kit
