Is there any standard protocols which is reproducible for detection of DNA on any form of electrodes. I see tons of papers on electrochemical detection of DNA but does someone have any experience reproducing the results from any paper? I mean I have seen people using PBS, acetate buffer and also mediators like ruthenium & methylene blue. Anyone have any experience with getting any of these?

In a sense, Electrochemical DNA sensing is a broad analytical bioapplication, including in general, labeled (indirect) or label-free(direct) techniques. In my experience, stripping voltammetry was one of the powerful, reliable, and reproducible approaches for ssDNA analysis.

The answer is: there is no any standard electrochemical methods for DNA detection. Amperometry and electrochemical impedance spectroscopy (EIS) have been widely used. However, the uncertainty is still pretty high if you hope the detection sensitivity down to (missing) single-base pair. It is hard to believe that electrochemical methods can detect DNA with the sensitivity at the single-base pair level, although a whole bunch of papers have claimed so. In my opinion, fluorescent spectroscopy is argurably a much better and more effecient way than electrochemistry for DNA detection.

the most sensitive electrochemical determination of DNA fagments is based on a hybridization process. Detection DNA was described also by E. Palecek and J. Labuda and A. Merkoci.
