I am very new to the field of AD. I was searching for a few candidate genes in the literature that I can check by qPCR and immunoblot to show Microglia activation from microglia culture.
I have these candidates;
1. for qPCR- TNF-a, IL-1b, Ccl3, iNOS, Cox2 (& GAPDH as control).
2. for immunoblot- TNF-a, iNOS, Cox2
Any suggestions of genes or papers will be very helpful for me at this moment.

Hi,
It seems like you are on the right track--- though do keep in mind that the term "activation" applies to a wide variety of phenotypes microglia show. The activation state is context dependent (LPS vs. IL-4 addition will induce different activation states, for example). I have attached three papers of interest (all related to single cell sequencing--- which is the gold standard technique for understanding microglia heterogeneity), one applying to an AD model in mice. In the AD related paper (Keren Shaul et al.) the authors describe a novel microglia subset known as "Disease Associated Microglia" (DAM). In addition to this, there are a variety of other subsets that arise during development and disease states (see Hammond et. al 2019). I hope this helps!
