Thermal shift assay is very new to me so please bear with me.

This is a very specific question and it does seem like a lot of people use BioRad CFX96 to do their Thermal Shift Assay using SYPRO Orange as the dye.

What is everyone using as their channel/filter when trying to detect SYPRO Orange, more specifically on their BioRad CFX96? I have tried selecting All Channels (in Scan Mode) and it didn't give me results that I have hoped for. Thus, I tried using FRET (as I've seen online, many people are using this but didn't specifically say which instrument they're using) on BSA and H2O as positive and negative. The curve on the BSA is weird...it fluoresces almost immediately without a "lag" phase. H2O on the other hand gave me high fluorescence but then it significantly decreases when the temperature increases. Should this not be low from the get go?

Hopefully someone can shed some light and help a friend out!
