Hi Mariana,

Which Antibody are you using? I found this antibody: http://www.cellsignal.com/products/primary-antibodies/caspase-8-1c12-mouse-mab/9746.

This one detects full length (57kDa), intermediate (p43/p41) and the p18 active fragment (18kDa). It does not detect the 10/12 band but the 12/10kDa fractions your antibody detects should be the small subunit (p10) which binds to the larger subunit (p18).  Caspase 8 is active when it is a heterodimer of p18 + p10 subunits, so I think you should analyse the 18 kDa and 10 kDa bands but I am by no means an expert in caspases so you should probably wait till someone else answers as well.

Caspase-8 cleavage fragment p26  generation is required for induction of robust apoptosis. This can be detected by Asp-391/374 antibody (see data+cat# in my paper below

However, generation of p10 is required to definitely kill the cells after apoptosis...otherwise the apoptotic cells may enter cellular transformation and survive. See my paper below (Figure 4c). This fragment also detected by same antibody (Asp-391/374)
