Dear everyone,

Recently, i am exploring the up-stream regulatory mechanism of a specific oncogene and speculate that this oncogene is pre-transcriptionally-mediated by DNA promoter metahylation. However, when i use the “Meth-primer-disign” websites to design the primer for detecting and verfying the DNA methylation status. I found that the predicted CpG island seems not to be located on the promoter region but stretch across the first EXON region and the starting zone of transcript. And the predicted and designed MSP primer seems to be located on the promoter region, but there is no CpG island there. I am wondering whether promoter region without CpG island could still be methylated and whether the predicted primer could be used for MSP analysis?
