We are going to transfect cell lines with lentivirus to induce an overexpression of a modified protein. The cell lines harbour the wild type of that protein and I would like to see if there are differences in the downstream signaling, using western blot analysis. Therefore I’m not sure if it is sufficient to use just the bulk of the transfected cells or if it is better doing single cell selection to find the highest overexpression.
