I'm currently working with MLR using murine cells (naive T co-cultured with BMDC). Should I add IL-2 along with other cytokines at the beginning, or add it later? Also, what concentration do you recommend?

I think, you do not need to add IL-2 into primary MLR.The adding may be helpful on restimulations if you plan to grow cell lines. In this case up to 5 IU/ml will be sufficient.
