When is the best time to add compounds to primary cortical neurons?

I have heard that primary cortical neurons mature at DIV12-14. Is this true and what do people think? Has anyone added compounds to neurons younger than DIV12?

I am treating neurons with compounds to assess their IC-50 activity.

Hi Joanna,

I have often worked with primary hippocampal neurons (from rats embryos). Neurons were grown for 7 days. Then I removed medium (containing serum-BSA) to add fresh medium without serum. And at DIV 7-10, I added my compounds. This worked well for me, and our lab published with this protocol.
