Do you mean to say that you're using two solvent; pet eth & meOH?

If so, pet. eth will be on top layer, most of lipids/fat compound dissolved in pet. ether will be on top.

I assume that you dissolved methanolic extract in acidified water and now want to extract by petroleum ether. If this is true then upper layer is petroleum ether and lower layer is acidic water.
