I'm planning to collect plasma from the tail of anesthetized mice. I am looking for the best method to determine corticosterone and ACTH concentrations in these samples ?
For corticosterone, I am always using the crystal chem ELISA kits #80556. However, be careful as anesthesia will impact corticosterone measurements compared to awake animals

Would the anesthesia impact corticosterone more than handling and stressing the mice to get a blood sample ? What protocol do you follow for blood sampling ?
i can’t remember if I did ACTH. Yes anesthesia will impact more than a wake measurements. For blood sampling I use the tail nick and a collection below 2 minutes. There is no need to restrain the animal (mouse or rat) and it has been shown to give stable results. If your animals have catheters you could use that as well though.
