I have a question regarding the cytometric analysis of ALPS.

In the international consensus, the elevation of double negative T cells (CD3+CD4-/CD8-) TCR alpha beta positive appears as required criteria. My question is: in the sequential gating strategy, where do I select the positivity percentage from?

For example, when I select the double negative (TDN) CD3+ lymphocytes it is 5% and of them when I choose the alpha-beta it gives me 13.7%. but in reality, this 15% is from the DNT subset, while that 15% represents only 0.7% of the total CD3+ T.
