You can cleave the ester bridge (pH 3 phosphate buffer) so that only the Chlorhexidine appears in an HPLC chromatogram.

I don't have a particular method for Chlorhexidine using pH 3 phosphate buffer, but it is very common. You can find the preparation of 0.05 M pH 3 phosphate buffer on the internet.
It is possible to use just UV spectrometry but it depends on the cleanliness and complexity of your sample matrix. Other interferences in your drug product may absorb but HPLC can separate them.
I had a similar problem with 'high potency' Vitamin A Acetate (millions of IU per g). The HPLC chromatogram was so clean you could just assay it with a UV absorbance (325 nm) and get a good number.
