I think you first need to do in vitro tests and, using IC50, you can determine the concentration you use to administer to the mice.
It's recommended to extract as much as possible to avoid multiple extraction which could lead to irreproducible results, if your work will be done after phytochemical investigation and in-vitro testing you should start using 100-200 gm of extract and mix thoroughly with the amount of ethanol that would cover the surface of the powdered plant material (300-600 ml) and repeat the process for 3 times at least, However if you intend to isolate active components from extract you should start at least with 3 KG of plant material so that you would get good yield of the extract.
