If the same plasmid with same insert is used for transformation into DH5 alpha (E.coli) and Stbl2 (E.coli) and if further done scale up. What will be the qualitative and quantitative difference in plasmid scaled up from DH5 alpha (E.coli) and Stbl2 (E.coli) ? Is it correlated with further in protein expression?

Both are cloning strains designed for improved transformation capacity and allowing efficient plasmid propagation. Therefore I don't think there would be any significant difference between the 2 about the yield for plasmid extraction. And no correlation can be made between plasmid prep and protein expression.

I am not entirely sure what do you mean by the term "qualitatively". But with regards to quantity, it is a property of the origin of replication in your plasmid which theoretically shouldn't deviate drastically given two cloning strains of E.coli

Here I have referred ''qualitative'' term for A260/280 and A260/230 ratio. Actually these ratios are also important in further protein expression studies.

Unwanted A260/280 and A260/230 ratios can be attributed to presence of proteins and/or reagents. I would suggest the miniprep kit by Qiagen since that has given me consistently good results.
