I am struggling to find published methodologies that describes the ratio of antifungal agent to Candida species, in order to determine and evaluate gene expression through transcriptional analysis. I will be using a novel medicinal plant with antimicrobial properties. I've determined the MIC of the plant using broth-microdilution, but now I would like to determine which genes are up-regulated or down-regulated, and I cannot find a suitable protocol which explains the volume of my plant extract needed to treat my Candida sample suspension with. If anyone could please assist me in this regard,, it would be highly appreciated!
Have you tried analysing your gene targets with the known MIC of your plant extract? I think that will be a good start. You can compare the expression of your gene targets with a known MIC of an antifungal drug.
