I have a compound that is readily soluble in DMSO for in vitro experiments (in cells), but I want to administer it daily in mice subcutaneously for a drug response study. I know undiluted DMSO is likely the wrong choice. What then is the best vehicle for that type of administration?

Hi Jonathan, I think you should try to see if this drug binds to serum albumin because it is the major carrier of drugs in the circulation.
If the drug does not bind serum albumin, it will have a very short half life in the circulation.
PBS/Saline is most ideal for in vivo applications. However, whether or not you can substitute DMSO with an alternate solvent would depend on your compound. The data sheet usually specifies which solvents the compound can be dissolved in. So if you have one on the list that is less toxic, you could use that. But keep in mind that different solvents may lead to different PK/PD. You would need to have a control group that is being injected with the same dose of the solvent (no drug).
