I am gonna perform electroporation in MKN45 GC cell lines, any suggestions for the suitable volts for this cell line ? Already tried normal transfection.
Typically, field strengths of 400 to 1000 V/cm are used for electroporation of mammalian cells which is usually performed in a buffered saline solution or culture medium with a single pulse ranging from 5 to 25 ms for DNA electroporation. For small molecules, such as siRNA, high voltage microsecond pulse lengths may be needed.
Also, for a 2mm gap cuvette, the voltage may range from 120 to 200 volts, and for a 4mm gap cuvette, the voltage may range from 170 to 300 volts.
For instance, for MKN-45 GC cells, you may use 130v for 12.5 ms, if you are using Gene Pulser Xcell electroporation system (Bio-Rad) for siRNA transfection. Please see the article attached below.
