I am following Cawthon's, 2009 method for measuring telomere length. I want to know if its ok to take sperm DNA as standard DNA for the standard curve

Since sperm DNA are supposed to have longer telomere length than that of somatic cells, my T/S ratio are all below 1. Is this a problem, as I see in many papers they are between 1 and 2.

Are you trying to determine the effect of methylation and Shelterin protein expression on telomere length?

If so, you can use some type of multiple regression analysis. The response might be normal. If so, you can use Multiple Linear Regression. If your telomere length is a count variable, you can use Poisson Regression.
