I have modified a pH-responsive mesoporous silica nanoparticle with a diameter around (80-90nm), and I needed to study the cytotoxicity of this MSN on colon cancer cells.

the cells were seeded in 96-well plates in a cell culture medium, then in the treatment step, I tried to disperse MSN in a complete media (DMEM), but MSN particles were not dispersed at all. so, the results after the MTT assay were inaccurate.

Are there any possible suggestions to disperse it in another suitable solvent? (Knowing that my MSN substance disperses well in distilled water and DMSO)

In some studies, DMSO was used at a concentration of 1%. Is it DMSO with water, media, or PBS?

Is it possible to use distilled water only for dispersion and then dilute it with media then apply it on the cells?
