I am trying to purify a polar compound ( polarity analysed based on Rf value in DCM system ) ethyl acetate cell free extract of a marine fungus and before running a column , i want to figure out the best solvent system for separating my compound from impurities which are also polar mainly.

Its a small organic compound , only when i can purify it i will come to know about its nature and molecular composition. Its showing good results in the assays so i am trying to separate it further from impurities. As per literature, i tried DCM:Methanol (95:5), but could not get desired separation.
