I´m doing an emulsion of oil and water by stirring 25 mL of crude oil in 100 mL. I would like to measure the number and the size of the droplet. I´ve read in the literature you could take picture with a microscope and thereafter, analyses the picture to count and measure the droplet. But so far, no article give a reference for such software. Do anyone know a good software to do that?
A lot depends on the nature of your images. If the droplets are clearly separated and nicely resolved, you'll have no trouble analyzing the images with open source image processing systems such as (http://imagej.nih.gov/ij/) ImageJ or (http://fiji.sc/Fiji) Fiji.
If the droplets' images have strong diffraction rings, or if their images overlap substantially, then you'll have more work to do. In those cases, you might have to write your own analysis routines in a language such as matlab, IDL or python that lends itself to sophisticated image analysis.
Finally, you'll save yourself a lot of work if you can prepare your samples in such a way that you get good clear images of well-isolated droplets. If so, and if you want very precise values for the size, you might want to consider obtaining images with holographic video microscopy and analyzing them with some of the latest HVM codes, which also are freely available.

In addition to what Dr. Grier says, I would recommend his own code for particle tracking (http://www.physics.emory.edu/faculty/weeks//idl/index.html) that we recently used exactly for the purpose of oil droplets tracking (http://www.nature.com/nphoton/journal/v7/n10/abs/nphoton.2013.192.html).
