Ideal dose: Depends may depend on the strain, but I guess 65mg/kg ip single dose should be more than sufficient (IT HAS TO BE ABSOLUTELY FRESH)
Body Weight: 200 to 225g (if Wistar) should be ok
Sucrose feeding overnight is needed (via drinking water)
Vehicle: citrate buffer, 100mM pH 4.5

Know also that sometimes a rat will simply not become diabetic. I did histology on the pancreas from a rat that I had injected three times. His beta cells had robust staining for insulin. His cohort all became diabetic after the first injection, but he did not, and ended up being a cagemate because we could not use him for out project.
