For calling somatic several events could happen. One is LOH. For LOH what should be the minimum depth of coverage in the paired diseased tissue which is homozygous at this locus (blood is a confident heterozygote site with read depth say ref15 and alt18).
Again a second tagged query is, exactly when would we call a locus as a LOH(what should be the cut off minor allele frequency of Blood).
Assuming the diseased tissue is blood-perfused, you should also be accounting for sample impurities. For example, if purity is 80%, then there is roughly a 0.2/2=10% chance that a given read would still indicate heterozygosity at your true homozygous site. If you can make a good estimate of purity, you can work out the statistics that would indicate the required depth of coverage for a given confidence.
