In the intracellular MIC determination what should be the concentration of the amikacin and gentamicin for extracllular killing of mycobaterium tuberculosis, also what should be the treatment time, to avoid the penetration inside the macrophage.
For the similar type of study, but Salmonella typhimurium as the infectant, I used 80 uMolar Gentamicin for 30 minutes (I titrated this concentration from 15 minutes to 2 hours). It worked well for me.

200 ug/ml MBC and 20 ug/ml MIC is reported for amikacin to kill extracellular bacilli. generally people use 200 ug/ml for 1-2 hr then gives 2-3 washes with prewarmed PBS/RPMI.
