Hello there.
Has anyone used the BCA method to determine the protein content of chitosan? What solvent do you use to dissolve chitosan? Did you use 1% acetic acid or another solvent? I will be glad if you share your experience and knowledge.

The solubility of chitosan occurs in dilute acid medium by protonation of the amino groups of chitosan in the NH3+ form. The factors that influence the solubility of chitosan are the type and concentration of acid, the ionic strength of the medium, and the degree of acetylation of chitosan. The solubilization of chitosan can be done by adding organic or mineral acid. The pH of the chitosan solution is then lower than 6.5. The most used acids are acetic acid and hydrochloric acid at a concentration of 1% or 0.1 M.
