What may be the possible reasons for getting only a single band during plasmid isolation?

Plasmids are always present in multiple forms. What might be the issue with my strain?

It can depend upon the plasmid and strain, it is possible to get a plasmid stock that is predominantly a single form, especially if you are using a RecA- strain that will stop formation of dimers and higher order recombination products.
