I am working with seawater samples and I am trying to collect the viruses from it.
I am new in this field and I've been having some trouble finding all the information I need.
So far, I used the Iron Chloride Precipitation of Viruses from Seawater protocol, from the Sullivan Lab, to filtrate the water sample and resuspend the viruses. At the moment I have collected the filters were the iron chloride and the viruses are trapped. Then I placed the filters in different 50ml tubes, and mixed them with a 0.1M EDTA-0.2M NaCl2-0.2M Ascorbate Buffer, pH6.5
After this step, I should end up with just viral DNA.
Has anyone done this procedure before? What protocol would you use to extract viral DNA after performing the Iron Chloride Precipitation?
I have been looking at other protocols and they mentioned to use the Wizard column, but I am getting confused if the column is used just to purify or also for DNA extraction
