I agree with Adam, you can also have a look at the Schwanhäusser Paper from 2011, they have a very good explaination on that topic.

Detecting/quantifying the target protein is the confirmatory step for the activity/effect of your drug. But, it also depends on whether you have the facilities and expertise to conduct protein level studies. mRNA level studies are much easier to conduct, as compared to protein level studies.
