Dear collegues. We are experiencing problems with induction of some of the rats. They tend tp wake up 10-15 seconds after they are taken out, even after 10 minutes in the chamber. What could be the problem? High metabolizers? Low chamber flow (1 l/min)? Low concentration of sevo (8%)? Thank you in advance.
Halothane which is still used by vets( low costs) is a very good gas to induce prolonged anaesthesia in rats. Sevo is too modern ! It's a gas for human
I used for induction 5% sevoflurane, N2O (60%), O2 (40%) with specific facial mask.
Maintains of anesthesia (4% sevoflurane, N2O 60%, O2 40%) - spontaneous breathing via facial mask for Rats.
Flow 100 - 200 ml/min.

Volatile anesthetics are rapid onset and recovery, mostly based on their lipid solubility. For sevoflurane, it is almost no metabolizing in vivo and just expired by respiration. Generally, the recovery from sevoflurane would be rapid even if longer exposure, the accumulation of sevoflurane in vivo is low. I don't know the purpose of your experiment, if you want longer anesthesia by sevoflurane, maintenance should be given rather than taking them out. Facial mask might be work. Good luck.
