I have read literature showed that the gelatin coating is around 50 nm thick. However, I am concerning whether the length of incubation time of 0.1% gelatin solution would change the thickness of gelatin coating. Also, would thicker gelatin coating favor cell binding?
Gelatin is a polymer dissolved in water. Gelatin films are typically highly hydrated, and therefore strongly dependent on the relative humidity to which they are exposed. Thus, any film thickness will be dependent on the water content of the polymer film at the time of measurement.
The age of the polymer in solution should not alter its final film properties, provided that the solution does not contain any ingredients that either degrade the polymer, or increase it's molecular weight or cause grafting or crosslinking to occur. In this context, pH would be quite important, since gelatin can be degraded at either very high or very low pH's.
