I have to calculate enzyme concentration for cellulase from Aspergillus niger based on a paper using cellulase from Trichoderma reesei ATCC26921. The provider for A. niger cellulase gave enzyme activity based on CMC (carboxymethylcellulose) while the producer for T. reesei ATCC26921 report enzyme activity with microcrystalline cellulose. I do not know how are they different and was not able to find an answer in published articles. Please help! Thank you all.
In both the case you measure the cellulase activity but you use two different substrates (microcrystalline and CMC).
Enzymatic Specific Activity (AS) corresponds to micromoles of products appeared per minute and per mg of protein. It is also expressed in the form: U / mg.
It can be calculated from the Enzymatic Activity (micromoles of producr formed per minute and per mL of protein, ie U / mL) determined experimentally and the concentration of protein in solution (mg / mL):
AS = AE / C

CMC hydrolysis is specific for endoglucanase activity and says very little about the ability of a cellulase preparation to hydrolyze microcystalline cellulose, which requires the synergistic action of endoglucanases, cellobiohydrolases, and beta-glucosidases
In one my work, we analysed the FPase, CMCase and b-glucodasidase of the fungi grew on PDA medium plus microcrystalline cellulose.
