Can anyone comment on the size on phospho Axl (Y779) protein size in HCC827 cells? I find that the unphosphorylated Axl full length protein is around 130 kDa in these cells, however, the phospho Axl usually gives 2 bands - (1) between 100 and 130 and (2) a stronger band between 130 and 170. I am using the same P-Axl antibody in PC9. I can see 3 strong bands- one above 100, one at 130 and one around 170 kDa. I have tried to IP with total axl (M-20 from SCBT) and blot with this abntibody to check P-Axl, and now i see one band right at 130 and one in between 130 and 170! I have blotted the IP-blot with P-Tyr (PY20) and the band at 130kDa doesn't come as P-Tyr!
Thank you for your comment Debjani. In my subsequent experiments I have analyzed some HCC827 drug resistant cells where I do see a single band for p-Axl using the same antibody slightly above 130. Hopefully this is the right band.
