A micro-organism might have antimicrobial activity against other organisms due to production of different metabolites, acids and enzymes etc. I want to check antimicrobial activity of some organisms focusing on their activity due to bacteriocins. Following study involves three methods to screen antagonistic activity of an organism; 1)Cross-streak method 2) Stab-Overlay method 3)Agar Well Diffusion method. I have read some other methods in other articles i.e; Agar spot method, Spot on lawn method, double plate method etc. I have understood that antimicrobial activity of bacteriocins are evaluated by testing supernatants after centrifugation cultures in broth.

Kindly guide me which method is the most suitable if someone has to check antimicrobial activity of an organism due to production of every type of antimicrobial compound,collectively by it?

Secondly, in given article, did authors evaluated this using first two methods i.e; 1)Cross-streak method 2) Stab-Overlay method?
