I am trying to isolate some halophilic streptomyces, but it doesn't work well.
I use Starch Casein Agar with different amount of NaCl up to 2% yet. Different types of microorganisms, including some bacillus grow on my plates.
Is there any way to make a selective media for halophilic streptomyces?
I add 1 g of CaCo3 to 10g of some soil samples today in order to reduce other microorganisms, will it work? Please help me find the best media for the isolation.

1. My best suggestion to isolate halophilic streptomyces medium was Starch casein agar with sea water and also Supplemented with Actidione 20 mg/l and Nalidixic acid 100 mg/l were added to minimize the other fungal and bacterial growth respectively. You can try this protocol it will give good results to you to get good number of actinomycetes or streptomyces.
2. Another suggestion better to get halophilic you can give the different concentration of salinity treatment to the soil samples afterwards use again same protocol what am mentioned above. So try this you will get good results.
Try the following medium:
1 g Beef extract
1g Yeastextract
2g Casein digest
15g Agar
Dissolve in 900mL seawater or diluent (membrane filtered water from the sampling area). Sterilize and add 100ml Nystatin sugar solution. Pour in plates.
Nystatin sugar solution
0.5g glucose
0.5g maltose
5mg nystatin
Dissolve in 100ml membrane-filtered diluent
