Depends on the abundance of the protein, affinity of the antibody and the strength of the fluorophore. I would start optimizing between 2 and 50 ng.
If you are procuring the antibody from suppliers like BD or eBiosciences etc, they will mention the required amount of antibody needed to be added per test in data sheet. Most of the time it works.
If you followed that and still if you are not getting the appropriate results then the permeabilizing step needed to be checked, like the composition of permeabilizing buffer or the incubation time, number of washing steps.( perm buffer should be used for dilluting antibodies and washing the cells)
Of course like Mr. Hendrikk Gremmels said it depends on the expression of protein itself and also if the supplier did not provide then the concentration it needed to be standardized with different concentrations.
