When I started to study about cell line culture I had a doubt that is why we should using the medium containing serum after when cells start coming off from the culture bottle? What material contains in medium containing serum for cell line?

Hi Bhaskara.
FBS contains many extra-cellular matrix (ECM) proteins such as Fibronectin.
These are required by the cells to stick to tissue-culture plastic (i.e. the cells DO NOT stick to the plastic, proteins in the serum stick to the plastic and the cells stick to the proteins).
If you use serum free media you have to coat the plastic with an ECM protein before adding cells (i.e. collagen, Fibronectin etc).
