I want the detailed information and pharmacology of how a statin causing Insulin Resistance as well as risk of new onset of T2DM.
Recent studies shows that Atorvastatin causes Insulin resistance while Rosuvastatin not. So, how Atorvastatin is causing the Insulin Resistance?

We discussed already in other circles here on research Gate the topic of diabetes and statins. Indeed there is a class effect but also a dose dependent and type of statins (hydrosoluble versus liposoluble) may hava an impact.

All statins increase the insulin resistance. This is a class effect. The degree to which a statin impairs insulin sensitivity is proportional to its LDL-C lowering effect. As a general rule it is more for stronger statins and for higher doses.
Of note this side effect of statins is outweighed by far by the effect of LDL-C lowering on outcome.

Henriksbo et al. provide evidence that various statins increase IL-1β secretion from macrophages through activation of the NLRP3 inflammasome.
The new study by Henriksbo et al. clearly shows that fluvastatin causes insulin resistance through activation of the NLRP3 inflammasome in murine adipose tissue explants. However, it remains to be seen whether statin therapy triggers the NLRP3 inflammasome in vivo and if it promotes insulin resistance in human adipose tissue. Future studies also should test whether statins induce inflammasome activation and insulin resistance in other metabolic tissues such as liver and skeletal muscle, as well as in pancreatic islets where NLRP3 inflammasome activation has been linked to enhanced IL-1β and T2D pathogenesis

Thats what i was also thinking about. The lipophilic or hydrophilic profile of statins. Atorvastatin is Lipophilic but Rosuvastatin is Hydrophilic. Again Rosuvastatin is way more powerful than Atorvastatin.
But Thierry C Gillebert stated that "The degree to which a statin impairs insulin sensitivity is proportional to its LDL-C lowering effect. As a general rule it is more for stronger statins and for higher doses." As per Thierry C Gillebert's statement Rosuvastatin will be more responsible for Insulin Resistance. But recent studies shows that Atorvastatin is more responsible for Insulin resistance. As it increases the Fasting plasma glucose level and glycated hemoglobin levels consistent with insulin resistance and increased ambient glycemia in hypercholesterolemic patients.
