the same inhibitors play different role in vitro and in vivo to block the activity of the protein . for example in in vivo study CsA donot block the activity well and NEM block completely and in vitro study NEM donot block the activity well and CsA block well.

You have to consider the network in vivo that is far more complicated than that in vitro conditions.
