I don't know what to do anymore. Everything I use is fresh, daily prepared. All solutions were remade, but I always get this effect. In the image is alpha-tubulin.
What is the voltage you have been using for running your gels and what are its dimensions? I only get this smile effect when the voltage is high and the gel runs too fast or it heats. If I were you would adjust the voltage recommended for the size of my gel or even run it under a lower voltage if you notice it is heating. Also check if the concentration of your running buffer is at maximum 1X.
check two aspects. 1. are the wells clean and straight when the gel is casted??make sure you rinse the wells with water after the comb is removed to clear traces of unpolymerized acrylamide and to check the shape and surface of the wells.
2. are the plates properly sealed on the sides? any air spaces at the sides will disturb the flow of current which can result in smiling effect of the bands. u can try sealing the plates at the sides with regular cello tape before fixing them and immersing in buffer.
I had this same effect some times and after changing all the buffers I discovered that it was because the acrylamide bottle had been in use for long time. Since then, I have a small aliquot approx 25-30 ml for use. Make sure that the acrylamide bottle has not been open for a long time, and as Dr. Vaidyanathan says, let the gel polymerize slowly overnight at 4 C, this also improves the resolution of the bands. .
