I am trying to find out the rate of glycerol consumption by Hansenula polymorpha at various combinations of stirring speed, air flow and even supplementing air with oxygen. With both modes of fermentation viz fed-batch and batch mode. Valuable inputs and suggestions are most welcome.
Your question is now some month old, but nevertheless you might still be interested, to get an answer.
First I would expect, that you will not find any influence of agitation or aeration as long as you avoid any oxygen limitation; but I would expect a difference between batch and (carbon limited) fed-batch mode.
The best approach to get a quantitative answer is this: plot biomass concentration vs. consumed amount of glycerol. For each cultivation phase where µ ist constant you will get a straight line. The slope of this line is the yield coefficient YX/S for glycerol. The rate of glycerol consumption is then QS [g/h] = µ * X * YX/S with µ = specific growth rate in 1/h and X as biomass (given as OD or dry matter g/l).
µ will be constant during batch phase and during fed-batch with exponential feeding and can be determined from same set of data.
