Hi, everyone. I have been conducting comet assay for analyzing the genotoxic effects of materials.
My problem is that the background of the microscope image is all green. So that I can`t observe my cells. Please tell me what should I do.

I have a feeling the problem lies in the filter. For SYBR green the excitation and emission are at 494nm and 521nm respectively. So please recheck the wavelengths. Comets are seen in the image.

Congratulations for receiving good results of your work. Seemingly, you used a high concentration of SYBR green leading to decreasing the quality of image(s). I strongly suggest that change the concentration of SYBR green through diluting in different densities. This is because, I experienced similar images (as yours) when I used ethidium bromide in Alkaline Comet Assay. However, things went well by changing the density of color.
