I conducted a plaque assay using the double agar overlay method for 46 hours, but wasn't sure if this was long enough for the phages to maximize their plaque frequency and diameter. The strain of phage used was T4r and the type of bacteria was E. Coli strain B. Is it possible that the number/diameter of plaques observed would have increased if they were incubated for a longer period of time? Some literature I have read has recommended a five day incubation period.
