Oxaliplatin (OxPt) has to be dissolved in a solvent to drug CRC cell lines to generate resistant cell lines. The dissolved OxPt would also be used in various other experiments. What is the most suitable solvent medium to dissolve OxPt to maintain its chemical properties and longer shelf-life? Where should dissolved OxPt be kept, -80C or -20C or 4C?

Oxaliplatin is soluble in DMSO up to 100mM. So you prepare a stock of oxaliplatin (100mM) in DMSO and store the stock at -20 degree C for long-term storage. Further dilutions of the stock solution using aqueous buffer like PBS (pH 7.2) or isotonic saline should be made prior to performing the experiment. Please note, do not store the diluted aqueous solution of oxaliplatin for more than a day. Use it for your experiment on that day and discard the rest.
While preparing dilutions of oxaliplatin to be used for in vitro experiment, prepare them in such a way that the DMSO concentration in the final solution to be added to the cells in culture should be less than 0.1%.
0.1% DMSO is considered to be safe for almost all cells.

Although Oxaliplatin is soluble in DMSO, the use of DMSO to dissolve Oxaliplatin in biological studies is strongly discouraged. The DMSO was found to insert itself into the ligand, resulting in loss of activity. (Reference Hall et al. (2014), Cancer research published online. PMID: 24812268).
Can oxaliplatin be dissolved in warm water to make the stock solution and stored in -20 degree C for long-term use? Further dilutions can be made with PBS.

I have similar question. We did order 200g of oxaliplatin powder. We want to solubilize this in DMF at 100mM. The solubility indicated on the MDS sheets means that we will not be possible given its 1.67 mg/mL solubility. What is the best way to proceed? Is weighing powder and recalculating a lower initial weight the best solution and what safety procedures do you recommend?

We found the best way to dissolve Oxaliplatin, i.e. milliQ water. You can keep the stock at -80 degree C for a longer time.
