I am trying to dilute the concentration of gold nanorod (AR 3.7) in order to adjust its longitudinal absorption peak at 0.25. But each time when I add water, after it reaches 0.5abs, the peak disappears. But the same gold nanorod coated by nanocarrier could be adjusted to 0.25. Is there any other protocol that I can follow?
An absorbance of 0.5 is a transmission of ~ 32%
An absorbance of 0.25 is a transmission of ~56%.
Detecting this range is not a problem with the spectrometer.
You are diluting by a factor of 2?
Yes. I am diluting by a factor of 2. I tried the same process second time as well, but unfortunately I can not see any longitudinal peak below than 0.5 abs.
The only thing I can think of is that there is some aggregation phenomenon happening at a critical concentration, and the spectral response changes dramatically below this. You could try diluting more gradually
