Can anyone please help me with the standard laboratory procedure for Lignin, Cellulose, Hemicellulose determination in organic manure along with the detail formula for calculation. I could see several modification from Standard Van Soest method. Please share with me ready laboratory method if anyone is using. It will be a great help.

If your sample is Biomass-based, you can use the method that have been published by National Renewable Energy Laboratory (NREL) with the title "Determination of Structural Carbohydrates and Lignin in Biomass". The preparation method uses a two-step acid hydrolysis to fractionate the biomass into forms that are more easily quantified. Please find the method in the attached file.
