We can get the mass of live cells by quantitative phase imaging, but we would also like to know the mass of total DNA within these cells. We wonder how reliable Hoechst 33342 labeling is on total DNA, since it only labels contiguous A-T base pairs. Can we use quantitative phase imaging to determine the total DNA mass in mammalian cells? If so, what is the best way to do so? If not, what other methods do you suggest?
