Hi Mennatallah,

RNA concentration for initial cDNA synthesis depends upon the variety of factors but 20-50ng/uL (DNA free) RNA is sufficient for the cDNA , then you can dilute your cDNA upto 4-5 times for all the samples according to the requirement (expression of house keeping gene(s) in your samples).

If you have low conc. of RNA that does not matter but careful in the step of RNA to cDNA conversion.In our lab, we have done RT using 1ng/ul conc. of RNA.
