Chromatin purification from paraffin-embedded sample for genome-wide epigenetic studies (pathology-tissue chromatin immunoprecipitation). What is the ideal protocol and the main factors that may interfere with the execution of the method?
I think chloroform/phenol method is the method of choice but find the best fit your samples. I modified one and get better DNA. Plus you need DNA that is not sheared. Let say most kits add more steps and time into extraction and that helps shearing your nucleic acid. My modified technique is here doi:10.1016/j.mimet.2016.07.019
